18 Venom

The main threat in Marvel's Spider-Man 2 is Venom, who is brought to life by the horror icon Tony Todd's truly menacing performance. The Venom symbiote had been teased in the post-credit scenes for both of the series' previous entries as a potential cure for Harry Osborn's illness.

Harry has been released from his treatment by the time the game's story begins, and claims to be cured as a result, wanting to use the symbiote to "heal the world." Unfortunately, somewhere this plan inevitably takes a turn for the worst as Peter bonds with the symbiote and abandons it upon realizing how much it is corrupting him, forcing it to bond with Harry once again to create the monstrous Venom.

17 Kraven

Kraven the Hunter brings his Great Hunt to New York City in Marvel's Spider-Man 2​​​​​​ after proclaiming his boredom with hunting animals and mortal humans, whom he deemed as "unworthy prey." One of his allies provided him with a tablet showing a map of New York and the various skilled, technologically enhanced, and superhuman beings that inhabit it, with the suggestion that they could finally give him the challenge he desires. Kraven and his team arrive in New York with various vehicles and equipment unleashing carnage across the city during their hunt, and shockingly murder several iconic characters from the Spider-Man mythos, setting them up as truly dangerous adversaries.

16 Lizard

As revealed in the Marvel's Spider-Man: Miles Morales post-credits sequence, Dr. Curt Connors has been working with Norman Osborn on the treatment for Harry, and plays a major role in Marvel's Spider-Man 2. Unfortunately, one of the targets of Kraven's great hunt is Dr. Curt Connors' reptilian alter-ego, The Lizard.

In Marvel's Spider-Man 2, Connors seems to be mutating into a far more monstrous creature than before, much as he does in the Amazing Spider-Man comics' Shed arc. This results in Peter and Miles trying to save Connors while protecting the city from him in an action-packed, lengthy chase sequence and boss battle that had been teased in the game's PlayStation Showcase gameplay footage.

15 Sandman

Sandman was teased in Marvel's Spider-Man through the game's backpack missions, as Peter had trapped him in a vial in their last encounter. As players look at the item, Peter can be heard wondering to himself if he would unleash the foe once more if he opened it. In Marvel's Spider-Man 2, Sandman does manage to get loose and wreaks havoc across New York in a memorable opening boss battle that sees both Peter and Miles use their abilities as they face a giant, skyscraper-tall version of the villain.

14 Wraith

Former police captain Yuri Watanabe's vigilante alter-ego, Wraith, appears in Marvel's Spider-Man 2, tracking down the mysterious cult known as "The Flame". Yuri was a great ally to Peter and was his main within the New York Police Department, but she spiraled and began taking the law into her own hands during Marvel's Spider-Man's The City That Never Sleeps DLC when her squad was ambushed and massacred by Hammerhead, and an off-the-books undercover sting failed, leading to the death of another of her officers.

13 Grizzly

A hunter who resembles Maxwell Markham's Grizzly, appears as part of Kraven's hunters, wielding a large axe in his showdown with Spider-Man. In the comics, Markham was a former wrestler who donned a grizzly bear suit and exoskeleton harness to take on Spider-Man, which definitely matches the hunter's look in the game. However, it's worth noting that Marvel's Spider-Man 2 never refers to this character by name, so it's unclear whether this is an official Grizzly appearance or merely an Easter egg.

12 Mysterio

The seemingly reformed Quentin Beck and his alter-ego Mysterio appear through a series of side-quests in Marvel's Spider-Man 2. After visiting his attraction at Coney Island, Miles can visit Beck's escape room-esque "Mysteriums" around New York to partake in a series of challenges, such as time-trials, to free the civilians trapped inside.

Mysterio's comic-accurate suit had been mimicked by an Empire State University student for a Halloween costume in Marvel's Spider-Man, but Beck sports a slightly more upgraded design in the sequel, eschewing his cape for a robed look that's closer to his Spider-Man: Shattered Dimensions style while projecting a menacing skull within his fishbowl helmet.

11 Scorpion

Several villains from previous Marvel's Spider-Man games appear following a mass breakout at the R.A.F.T. supermax prison, but some of them don't even come to blows with either webhead. Instead, they face Kraven during cutscenes as part of his hunt. One such showdown consists of a one-on-one fight between Scorpion and Kraven, with the former using huge mounds of sand to try to get the jump on the ruthless hunter.

10 Rhino, Shocker, Electro And Vulture

Other foes from the Marvel's Spider-Man series, Rhino, Shocker, Electro, and Vulture, all appear briefly or are teased in Marvel's Spider-Man 2 after being named targets for Kraven's Great Hunt. Unfortunately, none of these foes get the chance to face either Spider-Man in person during the game. Electro and Vulture are confirmed dead, with Electro's armor and Vulture's wings found alongside Kraven's recordings about how he bested them.

Both Electro and Vulture do appear alongside Rhino in a Mr. Negative-induced trip inside Peter's mind, but these appearances are brief. Shocker doesn't appear in person at all, which may be disappointing for fans who wanted to see a homage to the 90s Spider-Man cartoon with Shocker and the symbiote in Insomniac's universe. The name of Shocker's alter-ego, Herman Shultz, is spotted on a holding cell opposite Martin Li's in Kraven's lair, with the implication being that Kraven already made the two supervillains fight to the death.

9 Kingpin And Taskmaster

Despite Kingpin appearing in both previous Marvel's Spider-Man titles, he only appears in Marvel's Spider-Man 2 as an image on Kraven's list of potential targets. This is the same for Taskmaster, who doesn't return with his challenges for the web-heads to further study their abilities in the sequel. Taskmaster was defeated in Marvel's Spider-Man, but unlike the other foes in that game, he wasn't apprehended and vanished, leaving the possibility of his return open. Despite his absence in Marvel's Spider-Man 2, Taskmaster is still somewhere in Manhattan according to an icon on Kraven's map,
